If I had an XBox with a single Live account, 2 controllers, 2 headsets would
both players be able to communicate when playing live?

You'll need an additional XBL account for the other player.

In the same game??

No, it depends on the game.

Project gotham allows two users on the same xbox to play live. Comes up in
the room as the gamertag, and the gamertag (Guest).

The O.P. was asking if the other person would be able to use the
communicator, hence the requirement of an additional XBL account.

What I've been told is that the only way to have 2 players each with
operational communicators, play the same game, on the same console, is to
have 2 accounts/gamertags (one per player) and have one of the 2 load
their info into a memory unit.

Haven't verified this yet but would like to hear from others.

Yes, it's possible. Most games that allow for guests also allow for
multiple gamertags.
